3ml. of Sodium hydroxide soln.was added to 3 ml of supplied soln. & 2 drops of 1% CuSO4 reagent was added to it.

Absense of Proteins.

Molisch’s Test

 

 

2ml of sample solution is taken with 2 drops of 5% α-napthol or Thymol solution.

Carefully inclined the tube & poured

conc.H2SO4 drop wise, using a dropper,

along sides of the tube.

Violet colour

appeared at the

junction of two

liquids.

Presence of carbohydrate

is confirmed.

Iodine Test: (Indicator Test)

 

 

2 drops of dilute Iodine reagent was added to

3 ml of supplied solution.

A deep blue coloration appeared

Presence of Starch is confirmed.


 [N.B.~Starch is a complex carbohydrate (polysaccharide) produced by most green plants as a primary means of energy storage. It primarily exists in two forms: amylose (a linear, helical chain of glucose) and amylopectin (a branched chain of glucose). ]

3 ml of supplied solution.

Absence of Starch is confirmed.

Seliwanoff’s Test

 

 

1 ml. of supplied solution was added to 3 ml.of Seliwanoff’s reagent. Mixed thoroughly, boiled for 25 seconds and allowed to cool.

No red coloration in the solution.

Absence of  Sucrose.

Benedict’s Test:

 

 

5 drops of supplied solution was added to 3 ml Benedict’s qualitative reagent.The mixture was boiled for 2 minutes & cooled.

A brick red precipitate was obtained.

 

Presence of reducing sugar.

 

Fehling’s Test:

 

 

Solution"A" was mixed with solution"B" in 1:1. 2 ml of this mixture is added with few drops of the test sample to A test tube.Then it is placed in a water-bath at 60° C.

Red precipitate is obtained.

Presence of reducing sugar is confirmed.

Barfoed’s Test: (Confirmatory Test)

 

 

1 ml of supplied solution was added to 3 ml.of Barfoed’s reagent. The mixture was boiled for 30 sec and cooled.

 

No change.

Absence of Monosaccharides.

Presence of Maltose is confirmed.

Hydrolysis Test: (Confirmatory Test)

 

 

3 drops of Conc.H2SO4 was added to 5 ml of supplied soln.The mixture was boiled for 3 minutes. It was cooled and neutralized with solid Na2CO3 powder until no further effervescence occur.5 drops of this solution was added to 3 ml. of Benedict qualitative reagent. Mixture was boiled for 2 minutes and cooled.

A brick red precipitate was obtained.

 

Hence,presence of Sucrose is confirmed.

1ml. of supplied solution was added to 3 ml. of Seliwanoff’s reagent. Mixed thoroughly, boiled for 25 seconds and allowed to cool.

 

Red colour develops in the solution.

Presence of Keto Sugar.

Presence of Fructose is confirmed.
